Sp1与宫颈癌细胞放射敏感性的关系

摘要: 目的探讨Sp1 在不同种类宫颈癌细胞系的表达情况及其与宫颈癌放射敏感性的关系。方法通过蛋白免疫印迹 (Western Blot)和实时荧光定量PCR(RT-PCR)检测不同种类的6种人宫颈癌细胞系(SiHa、HeLa、Caski、Me180、Ms751、C33a) 中Sp1的基础表达水平,选择Sp1高表达细胞株和低表达细胞株,利用慢病毒载体进行SP1的沉默及过表达。采用克隆形成实 验检测沉默/过表达稳定株及其对照组经不同剂量X射线照射后的存活情况,曲线拟合计算不同剂量辐射后各放射生物学参 数。结果6种宫颈癌细胞Sp1表达量由高到低依次Me180>Caski>C33a>SiHa>Ms751>HeLa。Western blot证实成功构建稳定 沉默Sp1及过表达Sp1的宫颈癌细胞株。克隆形成实验结果显示,电离辐射作用后,靶向抑制Spl的表达可以使Me180细胞的 存活分数进一步降低,差异有统计学意义(P<0.05),同时,Me180/shSP1细胞SF2、D0、Dq值较对照组偏低,α/β增高。经X射线作 用后,稳定过表达Sp1(HeLa/SP1)细胞存活分数较对照组高,差异有统计学意义(P<0.05),HeLa/SP1细胞SF2、D0、Dq较对照组显 著升高,α/β值则显著降低。结论沉默Sp1的表达能够增加宫颈癌细胞的放射敏感性,而过表达Sp1则能增强宫颈癌细胞放疗 抵抗,Sp1在宫颈癌放疗过程中起着重要作用。

Abstract: Objective To investigate the role of specificity protein 1 (Sp1) in regulating radiosensitivity of cervical cancer cell lines. Methods We analyzed Sp1 expression in 6 different cervical cancer cell lines (SiHa, HeLa, Caski, Me180, Ms751, and C33a) using Western blotting and real-time PCR. Clonogenic survival assay and curve fitting were used to assess the changes in radiosensitivity of Me180 cells transfected with lentivirus-mediated shRNA vector targeting sp1 and HeLa cells transfected with sp1 over-expression vector. Results In the 6 cell lines tested, the cellular expression levels of Sp1 decreased gradually in the order of Me180, Caski, C33a, SiHa, Ms751, and HeLa. SP1 knockdown with lentivirus-mediated shRNA significantly lowered the survival rate of Me180 cells following radiation exposure (P<0.05), and obviously lowered the values of SF2, D0 and Dq but significantly increased α/β of the cells. Compared with the cells transfected with the mock vector, HeLa cells with sp1 over-expression showed a significantly increased survival following radiation exposure (P<0.05) with obviously increased values of SF2, D0 and Dq but significantly lowered α/β. Conclusion Silencing Sp1 can increase the radiosensitivity while Sp1 overexpression enhances the radioresistance of cervical cancer cell lines, suggesting an important role of Sp1 in radiotherapy for cervical cancer.
