肿瘤坏死因子-α对骨髓源性肥大细胞表达MMP-3、MMP-9、IL-17的影响

mouse bone marrow-derived mast cells in vitro

摘要: 目的肿瘤坏死因子-α(TNF-α)对骨髓源性肥大细胞(BMMCs)分泌MMP-3、MMP-9、IL-17的影响。方法原代培养小鼠
BMMCs,将培养8 周的BMMCs种植于12 孔板中。细胞分为4 个组:BMMCs+PBS组(对照组)、BMMCs+2 ng/mL TNF-α组
(2 ng/mL组)、BMMCs+10 ng/mL TNF-α组(10 ng/mL组)、BMMCs+50 ng/mL TNF-α组(50 ng/mL组),不同浓度TNF-α与BMMCs
作用后,在12、24 h收集细胞,用实时荧光定量逆转录聚合酶链式反应(real-time polymerase chain reaction, real-time PCR),在
mRNA水平检测MMP-3、MMP-9、IL-17 的表达。结果培养12 h 后,2 ng/mL 组、10 ng/mL 组、50 ng/mL 组MMP-3、MMP-9、
IL-17 mRNA的表达均高于对照组(P<0.05),并且随着TNF-α浓度的上升mRNA的表达显著升高(P<0.05)。24 h结果与12 h结
果一致,24 h 与12 h 之间比较,除50 ng/mL 组MMP-3 mRNA的表达无差异(P>0.05),其余各组之间MMP-3、MMP-9、IL-17
mRNA的表达显著升高(P<0.05)。结论TNF-α可以上调BMMCS表达MMP-3、MMP-9、IL-17,且具有浓度依赖性和时间依赖性。

Abstract: Objective To investigate the effect of tumor necrosis factor-α (TNF-α) on the release of matrix metalloproteinase-3
(MMP-3), MMP-9, and interleukin-17 (IL-17) in cultured mouse bone marrow-derived mast cells (BMMCs) in vitro. Methods
Primarily cultured mouse BMMCs at 8 weeks were exposed PBS (control) or TNF-α at the concentrations of 2, 10, or 50 ng/mL
for 12 or 24 h. Real-time PCR was performed to detect the mRNA expressions of MMP-3, MMP-9, and IL-17 in the exposed
cells. Results A 12-hour exposure of the BMMCs to TNF-α caused significantly increased expressions of MMP-3, MMP-9, and
IL-17 in a concentration-dependent manner (P<0.05). Prolonged exposures of the cells to 2 and 10 TNF-α for 24 h further
increased MMP-3, MMP-9, and IL-17 mRNA expressions, but exposure to 50 ng/mL TNF-α for 24 h increased only MMP-3 and
MMP-9 expressions but not IL-17 mRNA expression. Conclusions TNF-α treatment of primarily cultured BMMCs can
significantly increase the cellular expressions of MMP-3, MMP-9, and IL-17 mRNAin a time- and dose-dependent manner.
