吲哚美辛对K562细胞株BCR/ABL-Wnt/β-catenin信号通路的影响

摘要: 目的探讨吲哚美辛对K562细胞增殖的影响及其分子机制。方法采用不同浓度的吲哚美辛作用于K562细胞,MTT法
检测其对细胞增殖的影响,克隆形成实验检测吲哚美辛对K562细胞克隆形成能力的影响。RT-PCR分别检测bcr-abl、β-catenin
的mRNA表达变化,Western blot分别检测pBCR/ABL、总BCR/ABL、β-catenin、pGSK-3β、c-myc的蛋白表达变化。结果100、
200、400 μmol/L吲哚美辛作用细胞48 h后,以浓度依赖性方式抑制K562细胞的增殖和克隆形成能力。3种浓度的吲哚美辛处
理K562细胞后,pBCR/ABL、总BCR/ABL的蛋白表达依次减少,而bcr-abl mRNA水平没有明显变化。β-catenin mRNA和蛋白
水平依次减少;pGSK-3β、c-myc 的蛋白水平均明显降低。结论吲哚美辛可通过抑制BCR/ABL-Wnt/β-catenin 信号通路的活
性,从而抑制白血病K562细胞的增殖和克隆形成能力。

Abstract: Objective To investigate the effect of indomethacin on the proliferation and Wnt/β-catenin pathway in K562 cells.
Methods The cell growth of K562 cells treated with different concentrations of indomethacin was assessed with MTT assay,
and the colony-forming ability of the cells was evaluated by colony-forming assay. The mRNA expressions of BCR/ABL and
β-catenin were detected by RT-PCR, and the protein expressions of pBCR/ABL, total BCR/ABL, β-catenin, pGSK-3β and c-myc
were analyzed by Western blotting. Results Indomethacin significantly suppressed the growth and colony-forming ability of
K562 cells in a dose-dependent manner. Indomethacin treatment dose-dependently decreased the protein level of pBCR/ABL
and total BCR/ABL without affecting bcr-abl mRNA expressions. Compared with the control groups, indomethacin-treated
cells showed obviously decreased mRNA and protein expressions of β-catenin and decreased protein expressions of pGSK-3β
and c-myc. Conclusion Indomethacin inhibits the proliferation of K562 cells by suppressing the activity of bcr-abl-Wnt/
β-catenin pathway.
