Bim和细胞外调节蛋白在肝癌多药耐药细胞中的表达

摘要: 目的检测人肝癌耐药细胞HepG-2/ADM和亲本细胞HepG-2中ERK1,ERK2、ERK5和Bim 的表达,探讨其对肝癌细胞
多药耐药的影响。方法小剂量缓慢诱导法诱导建立人肝癌耐药细胞株HepG-2/ADM;CCK-8法测定HepG-2/ADM对多种化
疗药物的交叉耐药性;Western-blotting 检测MRP-1,P-gp,ERK1,ERK2,ERK5 和Bim 蛋白水平的表达;荧光定量PCR 检测
Bim mRNA的表达。结果化疗药物能够体外诱导肿瘤细胞产生耐药性,HepG-2/ADM 对ADM、5-FU和CDDP 的耐药指数分
别为6.8,4.1和4.5,且高表达MRP-1和P-gp蛋白;与亲本细胞HepG-2相比,HepG-2/ADM 中ERK1,ERK2和ERK5的表达均升
高,ERK1蛋白磷酸化水平无显著变化,ERK2磷酸化水平下降,且p-ERK1/2与ERK1/2的比值下降;Bim的mRNA和蛋白表达
均下降。结论细胞外调节蛋白激酶ERKs和Bcl-2家族的促凋亡蛋白Bim的表达与人肝癌多药耐药的发生密切相关。

Abstract: Objective To investigate the expressions of extracellular signal-regulated kinase (ERK) and the BH3-only subgroup
of Bcl-2 related proteins (Bim) in multidrug-resistant hepatocellular carcinoma (HCC) cells and their association with drug
resistance in the cells. Methods The multdrug-resistant HepG-2 cell line was established by treatment with gradually
increasing doses of ADM. CCK-8 assay was used to determine the drug sensitivity of the cells, and the expressions of MRP-1,
P-gp, ERK1, ERK2, ERK5, and Bim were detected with Western blot. Bim mRNA expression level was measured using
quantitative real-time PCR. Results The drug resistance indices to ADM, 5-FU and CDDP was 6.8, 4.10, and 4.5 in HepG-2/
ADM cells, respectively. The drug-resistantcells showed marked up-regulation of MRP-1, P-gp, ERK1, ERK2 and ERK5 with
down-regulated phosphorylated ERK2 protein expression but no significant changes in phosphorylated ERK1 protein
expression to result in a decreased ratio of P-ERK1/2 and P-ERK1/2. Bim mRNA and protein expressions were both decreased
in HepG-2/ADM cells. Conclusion ERK and Bim are related to multidrug resistance in HepG-2/ADM cells.
