大黄素对大鼠骨髓间充质干细胞增殖及造血因子表达的影响

摘要: 目的探究何首乌中主要成分之一的大黄素对骨髓间充质干细胞(BMSCs)的增殖、细胞周期、细胞凋亡的影响以及对造血
相关因子表达的影响。方法采用MTT法检测大黄素对BMSCs增殖作用,流式细胞仪检测大黄素对BMSCs细胞周期与细胞
凋亡的影响,并用RT-PCR方法探究大黄素对造血因子表达的影响。结果大黄素0.1 μg/ml、1 μg/ml在作用BMSCs 48 h与72 h
后增殖率明显增高(P<0.05);大黄素作用48 h 后,0.1 μg/ml 能促使细胞周期的G2/M期增多(P<0.05),1 μg/ml 使S期增多(P<
0.01),G1/G0期降低(P<0.05);大黄素在1 μg/ml作用48 h后能有效抑制细胞的凋亡(P<0.01);大黄素0.1 μg/ml、1 μg/ml在作用
BMSCs 48 h后对造血因子TPO的表达有显著提高作用(P<0.05)。结论大黄素具有促进BMSCs增殖作用,并且可能通过对
细胞周期与细胞凋亡造成影响达到增殖作用,并能促使BMSCs中造血因子TPO表达的升高。

Abstract: Objective To study the effect of emodin on the proliferation, cell cycle distribution, apoptosis and expression of
hematopoietic growth factors in bone marrow mesenchymal stem cells (BMSCs). Methods The proliferation of rat BMSCs
exposed to emodin was analyzed using MTT assay, and flow cytometry was used to detect the apoptosis and cell cycle
changes of the exposed cells. Real-time quantitative PCR was used to determine the mRNA expression of the hematopoietic
growth factors. Results Exposure to 0.1 and 1 μg/ml emodin for 48 and 72 h significantly enhanced the proliferation of BMSCs
(P<0.01). The cells exposed to 0.1 μg/ml emodin showed significantly increased percentage of cells in G2/M phase (P<0.05), and
1 μg/ml emodin exposure caused increased cells in S phase (P<0.01) and decreased cells in G1/G0 phase (P<0.05). Emodin
exposure for 48 h resulted in significantly decreased cell apoptosis (P<0.05). BMSCs treated with 0.1 μg/ml emodin showed a
significant increase in the expression of thrombopoietin mRNA (P<0.05). Conclusion Emodin can promote the proliferation of
BMSCs in vitro possibly by regulating the cell cycle distribution, cell apoptosis and thrombopoietin expression.
