非小细胞肺癌组织中CXCR4和Nrf2的表达及其临床意义

摘要: 目的通过检测临床非小细胞肺癌(NSCLC)标本中CXCR4和Nrf2的表达情况,以探究两者在疾病发生发展中的重要作
用及相关性,为肺癌诊断和治疗提供新的思路。方法应用免疫组化及Real-time PCR方法对资料统计完整的66例NSCLC标
本检测CXCR4及Nrf2的表达情况。结果在NSCLC组织中,CXCR4与Nrf2蛋白及mRNA的表达量均明显高于正常组织(P<
0.01)。CXCR4 的阳性表达与较大的肿瘤体积(P=0.048)、肿瘤低分化(P=0.024)、较晚的TNM分期(P=0.018)、淋巴转移(P=
0.004)及远处转移(P=0.016)相关。Nrf2的阳性表达与较大的肿瘤体积(P=0.008)、较晚的肿瘤TNM分期(P=0.028)、淋巴结转
移(P=0.038)与远处转移(P=0.023)相关。进一步发现NSCLC组织中CXCR4 与Nrf2 的表达呈正相关(r=0.324,P<0.01),且
CXCR4、Nrf2共同阳性表达与淋巴结转移及远处转移存在显著相关性。结论证明CXCR4和Nrf2的异常表达与NSCLC的进
展、恶性生物学行为密切相关,且二者在NSCLC的发生发展中存在协同作用。

Abstract: Objective To detect the expressions of CXCR4 and Nrf2 in non-small cell lung cancer (NSCLC) tissues and analyze
their association with the clinicopathological features of NSCLC. Methods We investigated the expressions of CXCR4 and Nrf2
in 66 NSCLC and corresponding distant normal tissue specimens using immumohistochemistry and real-time PCR. Results
The expressions of CXCR4 protein and mRNA were significantly higher in NSCLC tissue specimens than in the distant normal
tissues, while the expression of Nrf2 protein and mRNA increased significantly in NSCLC tissues compared to those in the
distant normal tissues (P<0.01). A high expression level of CXCR4 was positively correlated with a large tumor size (P=0.048),
poor differentiation (P=0.024), advanced TNM stage (P=0.018), lymph node metastasis (P=0.004), and distant metastasis (P=
0.016). The expression of Nrf2 protein was positively correlated with a large tumor size (P=0.008), advanced TNM stage (P=
0.028), lymph node metastasis (P=0.038), and distant metastasis (P=0.023). A strong correlation was found between CXCR4 and
Nrf2 expressions in NSCLC tissues (r=0.324, P<0.01), and the co-expression of CXCR4 and Nrf2 was strongly correlated with
lymph node metastasis and distant metastasis. Conclusion Abnormal expressions of CXCR4 and Nrf2 may contribute to the
progression and malignant biological behavior of NSCLC.
