整合素α9β1对大鼠角膜缝线术后新生血管形成及血管内皮生长因子A表达的影响

摘要: 目的研究整合素α9β1对大鼠角膜缝线术后新生血管(CNV)形成及血管内皮生长因子A(VEGF-A)表达的影响并探讨其
意义。方法39只SD大鼠,随机分为正常组(3只),对照组、α9β1组、α9β1抑制组各12只。角膜行缝线术后,对照组予以左氧氟
沙星点双眼;α9β1组予以左氧氟沙星+α9β1点双眼;α9β1抑制组予以左氧氟沙星+α9β1单抗Y9A2点双眼。术后每天在裂隙灯
下观察CNV生长情况,并于3、5、7、14d分别随机取3个实验组中各3只大鼠,在裂隙灯显微镜下行角膜照相,计算CNV面积;
RT-PCR、Western blotting法检测不同时间点角膜中VEGF-A的表达。结果3组大鼠缝线后均有CNV形成,在第7天CNV面积
达最大,对照组CNV面积为3.21±0.19;和对照组比较,α9β1组CNV明显增多为4.57±0.31(P<0.05);而α9β1抑制组CNV明显降
低,面积为2.03±0.26(P<0.05);第14天时CNV出现部分消退。RT-PCR和Western blotting检测结果示:正常角膜仅有少量
VEGF-A表达,角膜缝线术后,VEGF-A表达明显增加。在术后第7天CNV生长达峰值;与对照组比较,α9β1组VEGF-A表达明
显增高(P<0.05),而α9β1抑制组VEGF-A表达显著降低(P<0.05)。结论在角膜缝线术后,整合素α9β1可通过上调VEGF-A的
表达促进CNV生成;给予整合素α9β1抑制剂可下调VEGF-A的表达和显著减少CNV生成。

Abstract: ObjectiveTo investigate the effect of integrinα9β1 on corneal angiogenesis and the expression of vascular
endothelial growth factor A (VEGF-A) in rats after corneal suture.MethodsThirty-nine SD rats were randomly divided into
normal control group, corneal suture model group,α9β1group, andα9β1suppression group, and in the latter3groups, the
rats received topical administration of levofloxacin, levofloxacin + α9β1, and levofloxacin + anti-α9β1monoclonal antibody
(Y9A2) twice daily after corneal suture, respectively. At 3, 5, 7, 14days after the surgery,3rats were randomly selected from
each group to observe corneal neovascularization (CNV) under slit lamp. RT-PCR and Western blotting were used to detect the
expressions of VEGF-A.ResultsCNV was not observed in normal cornea. Neovascularization occurred in the corneal limbus
in the 3corneal suture groups, and the leak neovascularization count (CNVC), which occurred on the7th postoperative day,
was significantly increased afterα9β1treatment compared with that in the model group (4.57±0.31vs3.21±0.19, P<0.05) but
markedly decreased afterα9β1suppression (2.03±0.26, P<0.05). VEGF-A was expressed at a low level in normal cornea, and
increased significantly in the model group, reaching the peak level on7th day (P<0.05); corneal VEGF-A expression was further
enhanced byα9β1treatment (P<0.05) while significantly lowered byα9β1suppression (P<0.05).ConclusionIntegrinα9β1can
promote CNV by up-regulating VEGF-A expression andα9β1suppression produces the opposite effect after corneal suture in
rats.
