蛋白激酶CK2β在食管癌中的表达及其临床意义

摘要： 目的探讨蛋白激酶CK2β在食管癌组织中的表达及其在食管癌发生发展中的临床意义。方法采用免疫组化方法检测含
60对食管癌及其癌旁正常粘膜组织芯片中CK2β的表达水平。同时收集南方医院胸外科2012年2~4月期间手术切除的食管组
织标本10例，并提取蛋白和RNA，通过Western blot和实时荧光定量PCR的方法检测其CK2β的表达变化。结果CK2β在食管
癌组织中表达为：1.79%表达阴性，1.79%表达弱阳性，10.71%表达中阳性，85.71%表达强阳性；而正常粘膜组织96.67%表达阴
性，3.33%弱阳性。CK2β在肿瘤及癌旁正常组织中阳性表达的分布有显著差异（P<0.001）。CK2β蛋白在食管癌中的表达水平
与TNM分期显著相关（P=0.010）。Western blot及荧光定量PCR同样证实了食管癌组织中的蛋白激酶CK2β的表达水平明显升
高。结论蛋白激酶CK2β的表达与食管癌的发生发展及其恶性程度显著相关。

Abstract: ObjectiveTo investigate the expression of casein kinase2βinesophageal carcinomatissuesandanalyzeitsclinical
significance.MethodsThe expression of CK2βinatissuechipcontaining60normal esophageal mucosa and60colorectal
cancer specimens were detected immunohistochemically. Ten human esophageal carcinoma and adjacent normal tissues were
examined for the expression of CK2βproteinandmRNAusingWesternblottingandreal-timequantitativePCR, respectively.
ResultsA strong expression of CK2βwasfoundin85.71% of the esophageal cancer tissues;1.79% of the cancer tissues were
negative for CK2βexpression, and1.79% and10.71% of the cancer tissues were weakly and moderately positive, respectively.
In the normal mucosal tissues, 96.67% of the tissues were negative for CK2βand3.33% showed weak CK2βexpression,
showing a significant difference in the distribution of CK2βbetweennormal andesophageal carcinomatissues(P<0.001). The
expression level of CK2βinesophageal cancerswasassociatedwiththepathological stage(TNM)(P=0.010). Western blotting
and real-time quantitative PCR also confirmed an increased CK2βexpressionintheesophagealcancertissues. ConclusionThe
high expression of protein kinase CK2βiscloselyrelatedtothecarcinogenesisandmalignancyofesophagealcancer.
