知母皂苷B对大鼠海马注射β-AP(25~35)致tau蛋白磷酸化的影响

Effects of Saponin B from Anemarrhena asphodeloides Bunge on tau hyperphosphorylation induced by beta-amyloid peptide (25-35) in rats

摘要: 目的观察大鼠双侧海马注射β淀粉样肽[β-AP(25 ̄35)]后,海马内p53、Wnt途径抑制剂DKK和磷酸化tau蛋白表达变化,并用此动物模型观察知母皂苷B的作用。方法用RT-PCR法检测p53和DKKmRNA表达的变化,用免疫组化法检测磷酸化的tau蛋白。结果AD模型组海马内神经元的tau蛋白过度磷酸化,而p53和DKKmRNA的表达与假手术对照组相比也显著增多;而知母皂苷B给药治疗组均可使这种现象得到明显改善。结论知母皂苷B可通过抑制β-AP引起的p53及其后续的DKK的表达增加而有效改善AD大鼠脑内神经元的tau蛋白过度磷酸化。 

Abstract: Objective To investigate the changes in the expressions of p53, DKK (the inhibitor of Wnt pathway) and phosphorylated tau in rat bilateral hippocampus after β-amyloid peptide (β-AP) (25-35) injection, and observe the effect of saponin B from Anemarrhena asphodeloides Bunge (SAaB) in this model. Methods After bilateral injection of β-AP (25-35) into the hippocampus of rats, RT-PCR was performed for observing the changes in p53 and DKK mRNA expressions and immunochemistry carried out to detect the changes in phosphorylated tau protein. Results RT-PCR showed increased p53 and DKK mRNA expression and immunochemistry revealed increased phosphorylated tau-positive cells in rat hippocampus after β-AP (25-35) injection, and administration of SAaB significantly ameliorated these changes. Conclusion SAaB can significantly ameliorate β-AP-induced tau hyperphosphorylation by inhibiting increased p53 and DKK mRNA expressions in response to β-AP injection.
