Effects of macrophage migration inhibitory factor on collagenⅠ and Ⅲ mRNA expression in vascular smooth muscle cells in vitro

摘要: 目的 探讨巨噬细胞移动抑制因子(MIF)与血管平滑肌细胞(VSMC)合成胶原之间的可能联系。方法 取大鼠主动脉中膜平滑肌进行原代细胞培养,用第4代传代细胞分为两组,用25、50、75、100ng/ml的MIF刺激VSMC生长为实验组,未用刺激VSMC生长为实验组,采用RT-PCR,PCR方法测定VSMC胶原Ⅰ、ⅢmRNA含量。结果 与正常对照组相比,除25ng/ml外,其他3种不同浓度的MIF刺激SMC,胶原ⅠmRNA相对含量都显著增加(P<0.05)。100ng/ml的MIF刺激SMC,胶原ⅢmRNA相对含量较对照组显著增加(P<0.05),而其余各浓度胶原ⅢmRNA相对含量与对照组比较均无显著性差异。结论 MIF与VSMC合成胶原之间有关,MIF刺激可以促进体外培养的VSMC胶原Ⅰ、ⅢmRNA表达。

Abstract: Objective To investigate the association between macrophage migration inhibitory factor (MIF) and collagen synthesis in vascular smooth muscle cells (VSMCs). Method VSMCs isolated from SD rat aorta were cultured in different culture plates, and the fourth passage of cells were divided into two groups, one of which was simulated by MIF and the other served as control group. CollagenⅠand Ⅲ mRNA expression in the VSMCs was detected by reverse transcriptase (RT)-PCR and PCR. Result CollagenⅠand Ⅲ mRNA expression in MIF-treated VSMCs increased significantly as compared with that in the control cells (P<0.05). Conclusion MIF increases collagenⅠand Ⅲ mRNA expression of VSMCs in vitro, indicating its involvement in collagen synthesis in VSMCs.
