中国蝮蛇蛇毒中小肽SV-PP-3的分离及其抗血小板聚集作用

Purification of a small peptide SV-PP-3 from the venom of Agkistrodon blomhoffii siniticus and its inhibition of platelet aggregation

摘要: 目的 研究中国蝮蛇蛇毒中具有抑制血小板聚集作用的小肽。方法 采用低温离心、SephadexG-75凝胶层析、高效液相C-18反相层析从蛇毒中分离纯化小肽,并用血小板聚集仪测定分离的小肽在体外对磷酸腺苷(ADP)诱导的兔血小板聚集作用。结果 SV-PP-3是从中国蝮蛇蛇毒中分离出一种相对分子质量为1234.557的新的小肽。SV-PP-3在体外能显著抑制ADP诱导的兔血小板聚集,具有剂量依赖性。结论 SV-PP-3对ADP诱导的兔血小板聚集具有显著的抑制作用。

Abstract: Objective To investigate the inhibitory effect of the small peptide SV-PP-3 from the venom of Agkistrodon blomhoffii siniticus on platelet aggregation. Methods The small peptide SV-PP-3 was isolated and purified by centrifugation at low temperature, sephadex G-75 column chromatography and reverse-phase (HPLC) on C-18 column. The platelet aggregation induced by adenosine diphosphate (ADP) was measured by LBY-NJ aggregometer. Results SV-PP-3 was purified from the venom of Agkistrodon blomhoffii siniticus, which inhibited the platelet ADP-induced platelet aggregation in a dose-dependent manner. Conclusion SV-PP-3 can inhibit ADP-Induced platelet aggregation.
