糖基化终极产物对乳鼠心肌细胞胞内游离钙瞬间变化的影响

Transient cytosolic free calcium changes in cultured neonatal rat cardiac myocytes in response to advanced glycation end-product treatment

摘要: 目的 探讨糖基化终极产物(AGEs)刺激对体外培养乳鼠心肌细胞内游离钙的影响。方法 应用荧光钙离子指示剂Fluo-3/AM将体外培养3~5 d的乳鼠心肌细胞染色,激光共聚焦显微镜(LSCM)观察不同浓度AGEs(50、100、200 μg/ml)对心肌细胞瞬时刺激后细胞内游离钙离子浓度的影响。结果 与对照组相比,AGEs刺激可以迅速引起心肌细胞胞内游离钙浓度的升高,其恢复时程显著延长并呈剂量依赖性。结论 AGEs可能通过改变心肌细胞内游离钙离子浓度对心肌细胞造成损伤。

Abstract: Objective To investigate the effects of advanced glycation end-products (AGEs) on transient cytosolic free calcium in neonatal rat cardiac myocytes (CMs) cultured in vitro.Methods CMs cultured for 3 to 5 days in vitro were incubated with Ca2+-sensitive fluorescent indicator Fluo-3AM with light screening at 37℃ with 5% CO2 for 60 min. Changes of the fluorescence signal of free calcium caused by AGEs were measured under laser scanning confocal microscope (LSCM).Results Compared with the control cells, AGEs caused an increase in the concentration of cytosolic free calcium in a dose-dependent manner.Conclusion AGEs may impair neonatal rat CMs by altering cytosolic free calcium concentration.
