UrotensinⅡinduces hypertrophy of in vitro cultured neonatal rat cardiac myocytes

摘要/Abstract

摘要： 目的研究尾加压素Ⅱ(UⅡ)能否诱导体外培养的乳鼠心肌细胞肥大。方法心肌细胞体外培养40 h后,换无血清DMEM继续培养24 h,应用流式细胞仪和3H-亮氨酸(³H-Leu)掺入法观察不同浓度UⅡ作用24 h后对心肌细胞大小和蛋白掺入率的影响。结果 10^-7 mol/L的UⅡ能增加心肌细胞的大小(P=0.021)和³H-Leu掺入率(P=0.015)。结论 UⅡ能诱导体外培养的乳鼠心肌细胞肥大。

Abstract: Objective To examine whether urotensinⅡ(UⅡ) induces hypertrophy of neonatal rat cardiomyocytes cultured in vitro. Methods The primary cardiac myocytes cultured for 40 h followed by further culture in serum-free media for another 24 h were subjected to exposure to UⅡof varied concentrations for 24 h, after which the changes in the size of the cells were analyzed by flow cytometry with ³H-leucine incorporation also measured. Results At the concentration of 1×10^-7 mol/L, UⅡcould increase the size of the cultured cardiac myocardial cells (P=0.021) and ³H-leucine incorporation (P=0.015). Conclusion UⅡmay induce hypertrophy of neonatal rat cardiac myocytes cultured in vitro.
