预防性给予阿司匹林对大鼠中暑休克的保护作用

Pretreatment with aspirin for protection against heat stroke in rats

摘要: 目的 探讨中暑休克大鼠的血压及白介素-1β(IL-1β)水平变化及预先给予阿司匹林(ASA)可能具有的保护作用。方法 将Sprague-Dawley大鼠随机分为对照组及ASA处理组,每组10只。建立大鼠中暑休克模型,连续记录动脉压、结肠温度(Tco)、心电图(ECG)。另取对照组及ASA组大鼠(n=7)处理同前,测热暴露(HE)0及60 min时大鼠血浆IL-1β水平。结果 (1)两组大鼠平均动脉压在HE 0~50 min无显著差异,在55~60 min时开始下降,对照组血压较ASA组下降更显著(P<0.01),HE后K值及重搏切迹高度(hD/H)逐渐下降,尤其在HE 40 min后对照组K值较ASA组下降更为显著(P<0.05);(2)Tco在HE后升高,两组未见显著差异;(3)两组中暑休克形成时间无显著差异,ASA组生存时间较对照组显著延长(P<0.05);(4)ECG显示随着HE时间增加,两组心率、QT间期增加,PR间期降低;(5)HE后两组IL-1β水平均升高,对照组较ASA组在HE 60 min后IL-1β水平上升更高(P<0.05)。结论 预防性给予抗炎剂量阿司匹林可能对中暑休克出现的血压降低有一定的保护作用,这种作用可能是通过抑制IL-1β而实现的。

Abstract: Objective To observe the changes in blood pressure and interleukin-1β(IL-1β) level after heat stroke in rats and to observe the protective effect of pretreatment with aspirin against heat stroke. Methods Rat models of heat stroke were established and randomly assigned into control (n=10) and aspirin groups (n=10). Arterial blood pressure, colonic temperature (Tco), and electrocardiograph (ECG) were monitored and blood samples taken at 0 and 60 min after heat exposure for deter-mining the plasma IL-1βlevels in the two groups. Results From 0 to 50 min after heat exposure, the mean arterial blood pressure(MAP)was not significantly different between the two groups, but at about 55-60 min, the MAP significantly decreased in the control group in comparison with the aspirin group (P<0.01). The K value and the height of the dicrotic notch (hD/H) were gradually decreased, especially at 40 min after heat exposure, and the control group showed greater reduction in the K value. Tco of both groups were increased after heat exposure, without significant difference between groups. The time of the onset of heat stroke was similar in the two groups, but rats in the aspirin group had significantly longer survival time (P<0.05). ECG showed that the heart rate and QT intervals of both groups were increased, while PR intervals were decreased after heat exposure. Plasma IL-1βlevels in the two groups were significantly elevated at 60 min in comparison with the basal level (P<0.05), which was more obvious in the control group (P<0.05). Conclusion Pretreatment with anti-inflammatory dose of aspirin can provide protection against heat stroke in rats, which may be associated with the inhibition of elevation of plasma IL-1βlevels by aspirin.
