子宫内膜异位症患者肝细胞生长因子及其受体c-Met的表达

Expression of hepatocyte growth factor and its receptor c-met gene in the endometrium of women with endometriosis

摘要: 目的 研究肝细胞生长因子(HGF)及其受体c-Met在子宫内膜异位症发病机制中的作用。方法 采用mRNA原位杂交和免疫组织化学技术检测经腹腔镜手术及病理证实的54位子宫内膜异位症妇女(Ⅰ/Ⅱ期患者28例,Ⅲ/Ⅳ期患者26例)在位和异位内膜组织及24例正常对照组妇女在位内膜HGF、c-Met的分子和蛋白表达。结果 子宫内膜异位症患者在位内膜HGF、c-met基因和蛋白阳性表达与异位内膜一致,但阳性切片中,异位内膜阳性细胞数及表达强度高于在位内膜。增生期和分泌期的内膜细胞HGF/c-Met mRNA和蛋白的表达相近,无明显差异。对照组、Ⅰ/Ⅱ期患者在位、异位内膜与Ⅲ/Ⅳ患者在位、异位内膜比较,HGF、c-met基因和蛋白阳性表达频率依次上升,组间差异显著(P<0.01)。Ⅰ/Ⅱ期、Ⅲ/Ⅳ期患者在位和异位内膜与对照组比较,HGF、c-met基因和蛋白阳性表达频率有显著差异;Ⅰ/Ⅱ期患者与Ⅲ/Ⅳ期患者比较,HGF/c-Met阳性表达频率和表达强度均无显著差异。Ⅲ/Ⅳ期患者c-Met强阳性表达频率与对照组比较,有显著差异。结论 HGF、c-met基因和蛋白表达与子宫内膜异位症发病机制相关,在位内膜性质的改变可能是子宫内膜异位症发病机制的关键环节。

Abstract: Objective To study the role of hepatocyte growth factor (HGF) and its receptor (c-Met) in the pathogenesis of en-dometriosis. Methods In situ hybridization and immunohistochemistry method was used to examine HGF mRNA and protein expression in eutopic and ectopic endometrium obtained from 54 women in reproductive age with endometriosis confirmed by laparoscopy and histological examination (including 28 cases of stages Ⅰ/Ⅱand 26 of stages Ⅲ/Ⅳ) and from the endometri-um of 24 healthy fertile women. Results In patients with endometriosis, the positivity rate of HGF/c-Met expression was simi-lar in the eutopic and ectopic endometria, but the number of positive cells and expression intensity were much higher in ec-topic endometrium. The positive expression rate varied little between the proliferative and secretory stages of the endometri-um. Compare with the healthy controls, patients with endometriosis of stages Ⅰ/Ⅱand Ⅲ/Ⅳhad significantly elevated posi-tivity rate of HGF and c-Met expressions in either eutopic and ectopic endometrium, but without significant difference be-tween the two patient groups (P<0.01). The rate of strong c-Met protein expression was significantly higher in Ⅲ/Ⅳen-dometriosis than in the controls, without statistical difference between other groups. Conclusion The expressions of HGF and c-met gene are correlated with the pathogenesis of endometriosis, which is preceded by eventful changes in the biological be-havior of the eutopic endometrium.
