高效毛细管电泳法测定尿液中柠檬酸盐、草酸盐的含量

High-performance capillary electrophoresis for determination of urinary citrate and oxalate

摘要: 目的 以高效毛细管电泳法测定尿液中柠檬酸盐、草酸盐的含量。方法 以毛细管区带电泳分离分析尿样中的柠檬酸盐、草酸盐,以铬酸钠为紫外吸收剂进行间接紫外检测。结果 柠檬酸根、草酸根阴离子在5 min内得到很好的分离,迁移时间分别为3.76、3.14 min,柠檬酸根在4.00~40.00 μg/ml浓度范围内、草酸根在2.00~40.00 μg/ml浓度范围内线性关系良好,柠檬酸根阴离子最低检测限为2 μg/ml、草酸根阴离子的最低检测限为1 μg/ml。加样回收率分别为97.53%、99.11%。结论 该方法简单,准确,成本低,可为尿液中柠檬酸盐、草酸盐的直接测定提供参考。

Abstract: Objective To develop a method for determining urinary citrate and oxalate using high-performance capillary electrophoresis. Methods Capillary electrophoresis with indirect ultraviolet (UV) detection was performed for determining urinary citrate and oxalate. Sodium hromate was used as the UV-absorbing background electrolyte. Results Citrate and oxalate had a migration time of 3.76 min and 3.14 min respectively. The calibration curves were linear within a limit of about 2 μg/ml for citrate and about 1 μg/ml for oxalate. The recovery rates of citrate and oxalate were 97.53% and 99.11% respectively. Conclusion This method is simple, accurate and at low cost for determining citrate and oxalate in the urine.
