细胞周期蛋白依赖性激酶-2两种剪接体在SH-SY5Y细胞中共表达

Co-expression of two spliceosomes of cyclin-dependent kinase-2 in SH-SY5Y cells

摘要: 目的 观察细胞周期蛋白依赖性激酶-2(CDK-2)在SH-SY5Y细胞中的表达情况。方法 采用RT-PCR方法研究CDK-2的表达,用DNA银染技术观察PCR产物,割胶回收后,克隆到载体上、测序。结果 在聚丙烯酰胺胶上可以观察到两条DNA带,相对分子质量小的DNA带染色较弱;将两条DNA带切下,回收后与载体连接,测序证实两条DNA带均为CDK-2基因的产物。结论 CDK-2的两种剪接形式在SH-SY5Y细胞中共表达,其中一种剪接形式是第五外显子缺如,但其表达量比较弱,其作用机制仍有待于进一步研究。

Abstract: Objective To investigate the expression of cyclin-dependent kinase-2 (CDK-2) gene in SH-SY5Y cells. Methods The expression of CDK-2 gene was examined with reverse transcriptional (RT)-PCR, and the PCR products underwent electrophoresis on non-denaturing poly-acrylamide gel (PAG) followed by silver staining. The separated and purified DNAs were ligated into pMD18-T vector, and the positive clones identified by sequence analysis. Results Two DNA bands were displayed on PAG, and the one with smaller molecular weight was less intensively stained. The sequences of the two clones indicated that both were products of CDK-2 gene. Conclusion Two kinds of CDK-2 gene products are co-expressed in the SH-SY5Y cells, one of which lacks the fifth exon and is expressed at a low level.
