重组海葵肽类毒素hk2a对慢性充血性心力衰竭新西兰兔左心功能的影响

Effects of a novel recombinant polypeptide from the sea anemone Anthopleura sp. on left ventricular function in New Zealand rabbits with chronic congestive heart failure

摘要: 目的 观察重组海葵肽类毒素hk2a对慢性充血性心力衰竭(CCHF)新西兰白兔左心功能的影响。方法 采用腹主动脉缩窄术建立新西兰兔CCHF模型,利用超声心动图评价静脉注射hk2a前后不同时段兔左心功能的变化情况,并设立对照组进行比较。结果 hk2a能够在给药后即刻明显增加实验兔的左室射血分数(P<0.05),作用时间持续60 min以上(P<0.05)。与西地兰治疗相比具有起效快、作用强等特点。结论 新型海葵肽类毒素hk2a能够明显改善CCHF新西兰兔的左心功能。

Abstract: Objective To observe the effects of recombinant hk2a, a novel neurotoxin from the sea anemone Anthopleura sp. on the left ventricular function in New Zealand rabbits with chronic congestive heart failure(CCHF). Methods CHF model was established using New Zealand rabbits by abdominal aortic coarctation. The left ventricular ejection fraction (LVEF) was measured by Acuson ultrasound systems (Sequoia 512) at 0, 5, 15, 30, 45 and 60 min respectively after intravenous injection with a single dose of recombinant hk2a (20 μg/kg·b.w.). The response of the rabbits to hk2a treatment was observed in comparison with that of the control group. Results Recombinant hk2a immediately and significantly increased the left ventricular ejection fraction of the rabbit models of CCHF after intravenous injection (P<0.05), and its effects were maintained for more than 1 h without affecting the heart rate. Conclusion recombinant hk2a significantly increases the left ventricular ejection fraction in New Zealand rabbits with CCHF.
