2, 3, 5, 4'-tetrahydroxystilbene-2-O-β-d-glucoside alleviates lipopolysaccharide-induced acute lung injury in rats

摘要: 目的 观察2,3,5,4'-四羟基二苯乙烯-2-O-β-D-葡萄糖苷(TSG)对脂多糖(LPS)诱导的大鼠急性肺损伤(ALI)的保护作用,探讨其相关机制。方法 将36只SD大鼠随机分为正常对照组(n=9,蒸馏水 10 mL/kg,灌胃)、模型组(LPS组,n=9,脂多糖5 mg/kg,尾静脉注射)、TSG低剂量组(n=9, TSG 50 mg/kg,灌胃)和TSG高剂量组(n=9, TSG 100 mg/kg,灌胃)6 h后处死大鼠,观察大鼠肺组织病理切片并行肺损伤评分,测定肺组织湿/干质量比(W/D),ELISA法检测血清中炎症因子TNF-α和IL-6的浓度,同时检测肺组织中超氧化物歧化酶(SOD)的活力和丙二醛(MDA)的含量,并运用Western blot检测肺组织中NF-κB p65的表达水平。结果 与LPS组相比,TSG低、高剂量组肺病理损伤明显减轻(P<0.001),血清TNF-α和IL-6的浓度显著降低(P<0.001),肺组织W/D明显下降(P<0.001),SOD活性明显升高(P<0.001),MDA含量明显减少(P<0.001),NF-κB p65的表达水平显著下降,差异均具有统计学意义(P<0.001)。而TSG低剂量组和高剂量组之间比较并无差异(P 病理损伤评分=0.99,PTNF-α=0.99,PIL-6=0.86,PW/D=0.84, PSOD=0.76,PMDA=0.74,PNF-κB p65=0.17)。结论 TSG可减轻脂多糖诱导的急性肺损伤,其保护作用可能与抑制NF-κB通路的激活,提高抗氧化能力,减少炎症因子的释放有关。

Abstract: Objective To observe the protective effect of 2, 3, 5, 4'-tetrahydroxystilbene-2-O-β-d-glucoside (TSG) against lipopolysaccharide (LPS)-induced acute lung injury (ALI) in rats and explore the underlying mechanism. Methods Thirty-six SD rats were randomized equally into 4 groups: the normal control group, ALI model group, and low- and high-dose TSG groups (treated with 50 and 100 mg/kg TSG via intragastric administration, respectively). In all but the normal control group, the rats were subjected to tail vein injection of LPS to induced ALI. The rats were euthanized at 6 h after the injection for pathological examination of the lungs. The wet/dry weight ratio (W/D) of the lungs were calculated, and superoxide dismutase (SOD) activity and malondialdehyde (MDA) content in the lung tissues and serum levels of TNF-α and IL-6 were determined. Western blotting was performed to detect the levels of NF-κB p65 in the lungs. Results Compared with those in LPS group, the TSG-treated rats showed significantly milder lung pathologies (P<0.001) and had lower serum TNF-α and IL-6 levels (P<0.001) and W/D of the lung tissues (P<0.001), higher SOD activity (P<0.001) and lower MDA content in the lungs (P<0.001), and significantly lower expression of NF-κB p65 in the lungs (P<0.001). None of these indices showed significant differences between the low and high- dose TSG treatment groups (P>0.05). Conclusions TSG can ameliorate LPS- induced ALI in rats possibly by suppressing the NF-κB pathway to improve the antioxidant capacity and decrease the release of inflammatory factors.
